Reaching the age of 100 is unusual. Reaching that age while remaining relatively free from major age-related diseases is even more remarkable.

This is why centenarians and other exceptionally long-lived people have become an important focus of longevity research.

Scientists study these individuals to understand what may have helped them survive into very old age. Genetics is one part of that investigation.

Longevity tends to cluster in some families, suggesting that exceptionally long-lived people may inherit genetic characteristics that contribute to their ability to survive longer or delay certain age-related diseases.

Studies of long-lived populations have helped identify several genetic associations with longevity. Among the most consistently observed are variants involving APOE and FOXO3.

But centenarians do not all share one protective gene-or one genetic profile.

Instead, their genetics reveal a much more complex picture. Some people may carry combinations of variants that reduce susceptibility to particular diseases. Others may have genetic characteristics that support cellular maintenance, metabolism or resilience to biological stress.

This is an important distinction. Living to 100 may depend not only on genetic factors that directly support longevity, but also on avoiding or delaying diseases that commonly shorten life.

Studying centenarians can therefore help scientists identify biological pathways involved in both lifespan and healthspan.

Yet genes cannot fully explain exceptional longevity. Environment, lifestyle, healthcare and many other influences accumulate across an entire lifetime.

Centenarians are valuable not because they provide a simple genetic formula for living to 100, but because they offer a unique opportunity to understand how human biology can remain resilient for an exceptionally long time.

And what scientists learn from them may eventually help us understand healthy aging in everyone-not only those who reach 100.
